Campomanesia adamantium (Cambess.) O. Berg seed desiccation: influence on vigor and nucleic acids Anais da ABC (AABC)

The aim of this study was to evaluate the sensitivity of Campomanesia adamantium seeds to desiccation by drying in activated silica gel (fast) and under laboratory conditions (slow). To assess the sensitivity of the seeds to desiccation, we used drying with silica gel and drying under laboratory conditions (25 °C), in order to obtain seeds with moisture content of 45, 35, 30, 25, 20, 15, 10 and 5%. The physiological potential of the seeds after desiccation was evaluated by measuring primary root protrusion, percentage of normal seedlings, germination seed index, seedling length, total seedling dry mass, electrical conductivity and DNA and RNA integrities. Nucleic-acid based methods for bacterial identification are extremely useful in diagnostic applications due to their specificity and sensitivity. However, they require an optimal purification of the target molecules. As part of the development of a new diagnostic method for the detection of bacterial RNA in cow milk, we have compared four commercially available RNA extraction kits for the isolation of bacterial RNA from spiked UHT milk samples. Control of gene expression in Trypanosomatidae BJMBR

The study of mechanisms which control gene expression in trypanosomatids has developed at an increasing rate since 1989 when the first successful DNA transfection experiments were reported. Using primarily Trypanosoma brucei as a model, several groups have begun to elucidate the basic control mechanisms and to define the cellular factors involved in mRNA transcription, processing and translation in these parasites. This review focuses on the most recent studies regarding a subset of genes that are expressed differentially during the life cycle of three groups of parasites. Abstract Background Earthworms are sensitive to toxic chemicals present in the soil and so are useful indicator organisms for soil health. Eisenia fetida are commonly used in ecotoxicological studies; therefore the assembly of a baseline transcriptome is important for subsequent analyses exploring the impact of toxin exposure on genome wide gene expression. Development of genomic probes for detection of hepatitis A virus and enteroviruses in shellfish ArchiMer

The development of molecular biology techniques has modified research procedures for certain viruses, particularly hepatitis A virus (HAV) and enteroviruses. Accordingly, we designed cDNA and RNA genomic probes to detect viruses in shellfish in the environment. Better sensitivity was noted with RNA than cDNA probes: only 17/83 shellfish samples were positive with HAV and enterovirus DNA probes, where as 42/83 were positive with HAV and enterovirus RNA probes.

Abstract This study aimed to use gene chips to investigate differential gene expression profiles in the occurrence and development of acute myocardial infarction (AMI). The study included 12 AMI patients and 12 healthy individuals. Total mRNA of peripheral bloodwas extracted and reversed-transcribed to cDNA for microarray analysis. After establishing two pools with three subjects each (3 AMI patients and 3 healthy individuals), the remaining samples were used for RT-qPCR to confirm the microarray data. From the microarray results, seven genes were randomly selected for RT-qPCR. RT-qPCR results were analyzed by the 2-ΔΔCt method. This study was conducted to evaluate the effect of cottonseed meal supplemented with lysine and enzyme on laying hens performance and egg quality. Eighty Hy-Line W-36 white Leghorns were allotted for 12 weeks in a 2 × 2 factorial experiment in a completely randomized design, including four treatments and five replications with four birds in each. Treatments included: basal diet + 1% lysine + 0% enzyme; basal diet + 1% lysine + 0.025% enzyme; basal diet + 2% lysine + 0% enzyme; and basal diet + 2% lysine + 0.025% enzyme. In the liver of rainbow trout (Salmo gairdnerii ), vitellogenin (Vg) synthesis is influenced by oestradiol (E sub(2)) which is believed to act through the classical mechanism of steroid hormone action. After binding of the hormone to a soluble specific receptor protein, the oestradiol-receptor complex is translocated to the nucleus, where it interacts with DNA and modulates expression of Vg genes, leading to increased synthesis of specific mRNA and Vg. Response of Paenibacillus polymyxa to iron: alternations in cellular chemical composition and the production of fusaricidin type antimicrobial compounds BABT

In this work, growth, cellular chemical composition and production of fusaricidin type antimicrobial compounds by P. polymyxa SQR-21 were compared in tryptone broth supplemented with four concentrations of iron (25, 50, 100 and 200 µM). The data revealed that the growth of P. polymyxa SQR-21 was increased by 3-8% with the increase in concentration of ferric ion (Fe3+). The production of fusaricidin type compounds was increased by 33-49% only up to 50 µM Fe3+ and the highest level of Fe3+ was inhibitory. The effect of nucleic acids on growth, ureagenesis and nitrogen excretion of gilthead sea bream Sparus aurata juveniles ArchiMer

The aim of this trial was to evaluate the effect of dietary nucleic acids on ureagenesis and nitrogen balance in gilthead sea bream juveniles. For that purpose, 5 isonitrogenous (7.25% N) diets were formulated based on fish meal as protein source (FM-control diet) and to partially replace the fish meal nitrogen with two levels of nucleic acids (diets RNA1 and RNA2) or brewers yeast (diets BY1 and BY2). Each diet was fed for 10 weeks, to apparent visual satiety, to triplicate groups of 50 fish each with an average body weight of 12.7 g.
